Prazan 10 mg Tablet

Potassium-competitive acid blocker (P-CAB) — strong, fast acid suppression for:

• Erosive oesophagitis (healing and maintenance) and reflux (GERD)
• Gastric and duodenal ulcers
• Prevention of ulcers with low-dose aspirin or NSAIDs
• H. pylori eradication (with antibiotics)

Adults:

• Erosive oesophagitis: 20 mg once daily for 4 weeks (up to 8 weeks); maintenance 10–20 mg once daily.
• Heartburn of non-erosive GERD: 10 mg once daily for 4 weeks.
• Gastric ulcer: 20 mg once daily for 8 weeks; duodenal ulcer 20 mg daily for 6 weeks.
• Prevention with aspirin/NSAIDs: 10 mg once daily.
• H. pylori: 20 mg twice daily with amoxicillin (± clarithromycin) for 7–14 days.
• Take with or without food, at any time of day.
• Not for children.

• Allergy to vonoprazan
• Use with atazanavir or rilpivirine

Common: diarrhoea, constipation, nausea, abdominal discomfort, rash.

Less common: headache, raised liver enzymes, swelling.

Rare / with long-term use: C. difficile diarrhoea, severe skin reactions, liver injury, raised gastrin, possible low magnesium or B12 (as with PPIs).

• Exclude stomach cancer if alarm symptoms are present.
• Use the shortest duration appropriate; review long-term use.
• Severe liver or kidney impairment: caution.

Limited data; use in pregnancy only if benefit outweighs risk. Avoid breastfeeding during treatment.

• Atazanavir, rilpivirine (contraindicated)
• Drugs needing acid for absorption: ketoconazole, itraconazole, erlotinib, nelfinavir (reduced)
• Strong CYP3A4 inhibitors (clarithromycin) raise vonoprazan levels; inducers (rifampicin) lower them
• Digoxin, methyldigoxin (levels may rise)
