Prulose 2 mg Tablet

Selective 5-HT4 agonist (bowel motility stimulant) for:

• Chronic constipation in adults when laxatives have not given adequate relief

Adults: 2 mg once daily, with or without food, at any time of day.

Elderly over 65 and severe kidney impairment (eGFR < 30) or severe liver impairment: start 1 mg once daily (may increase to 2 mg in elderly if needed).

• Review after 4 weeks; stop if no benefit.
• Not for children.

• Dialysis-dependent kidney failure
• Bowel perforation or obstruction, obstructive ileus
• Severe inflammatory bowel disease (Crohn's, ulcerative colitis), toxic megacolon
• Allergy to prucalopride

Very common (mostly first day): headache, nausea, diarrhoea, abdominal pain.

Common: dizziness, tiredness, vomiting, wind, frequent urination.

Rare: palpitations; mood changes or suicidal thoughts (reported — seek help).

• Severe diarrhoea may reduce the effect of oral contraceptives — use additional contraception.
• Heart disease or arrhythmia history: caution.
• Report any low mood or suicidal thoughts.

Not recommended in pregnancy; use effective contraception. Not recommended while breastfeeding.

• Ketoconazole and other strong P-gp inhibitors (raise levels modestly)
• Atropine-like drugs (may reduce effect)

Headache, nausea, diarrhoea. Supportive care, fluids.

Store below 30°C, in the original pack.
